Measures Free Protein S in human plasma.Protein S is a Vitamin K dependent glycoprotein synthesized in the liver. In normal human plasma, Protein S is present in two forms: Free Protein S (40% of total Protein S concentration) and Protein S complexed with C4b binding protein (C4b-BP). Only Free Protein S is active and acts as a cofactor for activated Protein C in the inactivation of fVa and fVIIIa.The determination of both Free Protein S and Total Protein S aids in the differential diagnosis of three types of Protein S deficiencies:Type I - reduced levels of both Total and Free Protein S antigenType II - normal Total and Free Protein S antigen but reduced activityType III - normal Total Protein S levels but reduced Free Protein S levels. This ELISA is not reactive with Protein S-C4b-BP complexes.
